Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: 50mM HEPES, pH 7.4, 100mM NaCl, 2mM EDTA, 3mM DTT, 5mM PNPP substrate, 30 min at 30°C. Absorbance is measured at 405nM.

Background: Mammalian PTPases can be subdivided into 1 of 2 broad categories: transmembrane receptor PTPases and intracellular PTPases. PTPN11 is one of the 2 closely related mammalian intracellular PTPases whose sequences encode 2 tandem SRC homology 2 (SH2) domains that are located at the amino-terminal side of a single PTPase catalytic domain. This PTP is widely expressed in most tissues and plays a regulatory role in various cell signaling events that are important for a diversity of cell functions, such as mitogenic activation, metabolic control, transcription regulation, and cell migration.

Description: Human recombinant SHP-2, also known as PTPN11 (protein tyrosine phosphatase N11), full length (a.a. 2-593 (end) with N-terminal His-tag, Genbank accession number NM_002834, MW=70 kDa, expressed in an E. coli expression system.

Format: Aqueous buffer solution

Formulation: 40mM Tris pH 8.0, 110mM NaCl, 2.2mM KCl, 0.04% Tween-20 and 20% glycerol.

Storage Stability: At least 6 months at -80°C. Avoid freeze/thaw cycles. Protein may be diluted to ≥100 µg/ml in PBS + glycerol and stored at -80°C.
