Background: Sphingomyelin Synthase is a bidirectional lipid cholinephosphotransferases capable of converting phosphatidylcholine (PC) and ceramide to sphingomyelin (SM) and diacylglycerol (DAG) in addition to performing the inverse reaction. The direction is dependent on the relative concentrations of DAG and ceramide as phosphocholine acceptors. It directly and specifically recognizes the choline head group on the substrate. It also requires two fatty chains on the choline-P donor molecule in order to be recognized efficiently as a substrate. It may not function strictly as a SM synthase and is inhibited by bacterial PC-phospholipase C inhibitor D609. It is localized to Golgi apparatus; Golgi membrane; multi-pass membrane protein. It can be possibly present on the brain, heart, kidney, liver, muscle and stomach.
