TdT(Terminal-deoxymucleotidyl tranferase) is present in nuclei of immature T and B lymphocytes found in normal thymus and bone marrow. Some neoplastic cells of acute lymphoblastic lymphoma/leudemia have been shown to have high TdT activity. Mauter lymphocytes from normal peripheral blood usually do not contain the enzyme. Detection of nuclear expression of TdT by flow cytometry is a valuable technique in the characterization of leukemias and monitoring minimal residual leukemic cells.
