Assay Conditions: Enzyme reaction was conducted in abuffer (40 µl) containing 40 mM Tris (pH 7.4), 10 mM MgCl2, 2 mM MnCl2, 0.1 mg/ml BSA, 100 µM ATP, 100 µM PI:PS substrate (BPS catalog #40560) and 0.6-5 ng/µl Vps34 at 30°C for 20 minutes. Kinase reaction was measured using ADP-Glo reagents (Promega).
